    pdf format Publications Studies on the role of thyroid hormones action in the central nervous system Introduction Our research focuses on understanding the role of thyroid hormones T4 or thyroxine and T3 or 3 5 3 triiodothyronine action in the central nervous system CNS both during development and in adult stages We are especially interested in the identification of structural and or functional alterations in the CNS due to defects in thyroid hormones availability or signaling situations To achieve this goal we analyze the phenotype of experimental animals deficient in proteins involved in the metabolism and plasma membrane transport of thyroid hormones using different experimental approaches We perform in vivo studies to characterize the structural and functional consequences of situations with an abnormal availability of thyroid hormones in the CNS We also analyze the histopathology of human autopsy brain tissues from Allan Herndon Dudley syndrome patients This syndrome is due to mutations in the monocarboxylate transporter 8 MCT8 SLC16A2 an important thyroid hormones transporter in the brain Our studies will shed light on the pathophysiology and on the CNS disease mechanisms associated to defects in thyroid hormones signaling and will even contribute to the development of new therapeutic strategies These

    obesity studies Introduction Obesity is recognized as one of the most important public health problems facing the world today Prevalence is growing exponentially worldwide both in developed and developing countries An increased intake of energy dense foods especially when combined with reduced physical activity surely contributes to this high prevalence Obesity is defined medically as a state of increased adipose tissue of such magnitude to produce adverse health consequences Obese subjects have an increased risk of suffering insulin resistance type 2 diabetes and cardiovascular disease among others Previously considered an inert depot fat is now regarded as a key highly active and versatile organ with many important physiological and pathological roles Most of the adipose tissue functions are carried out via molecules secreted by fat cells capable of acting locally and in many non adipose tissues such as muscle liver and stomach tissues as well as nervous immune and vascular systems In humans the adipose tissue is mainly shown in two compartments the subcutaneous and the visceral fat depots Numerous methods have been developed to study the adipose tissue among them Proteomics Proteomics is the large scale study of proteins in disease related processes at the whole cell or tissue level and even in subcellular structures and in biological fluids We have used this approach in our laboratory and several comparative proteomic studies have been performed based on 2D DIGE and Mass Spectrometry currently considered pioneers in the field of obesity In a first study the subcutaneous and omental adipose tissue proteomes were compared highlighting the elevated metabolic activity and the higher inflammatory profile of the omental versus the subcutaneous fat in a second study the omental adipose tissue from obese and non obese subjects was compared underlining a clear distinctive proteomic pattern between the two group of individuals which

    a more academic than functional point of view macrophage activation can be classified as pro inflammatory M1 polarization anti inflammatory M2 or pro resolution deactivation M0 these profiles coexisting in the course of the immune response A line of interest has been the characterization of signaling pathways that determine the polarization and its effect on the release of mediators of inflammation In addition to this these mediators affect the function and gene expression in differentiated cells such as hepatocytes cardiomyocytes and myofibroblasts We recently analyzed the metabolic aspects associated with macrophage activation trying to answer the question about what changes in the regulation of energy metabolism and precursors NADPH riboses etc accompany the different types of polarization and to what extent these changes are necessary for the activation phenotype To get an idea of the magnitude of changes involved for example after M1 activation through TL4 challenge there is an alteration in the expression of over a thousand of genes The interest of these studies is to envisage the possibility to regulate macrophage function by altering their metabolic activity as a complementary strategy to regulate their participation in the inflammatory response We could show that regardless of the stimulus used and the availability of energy substrates the macrophage is in more than 90 glycolytic with limited use of other fuels for energy purposes however the pathways to generate metabolites from the TCA and glutaminolysis are fully functional and these molecules are used for other purposes The other area of interest in the group is the study of the role of macrophages that infiltrate the tissues under pathophysiological situations Following an ischemic process or in the course of infectious myocarditis the heart elicits an inflammatory response that promotes the infiltration and activation of resident and circulating inflammatory cells representing up to
