    on the study of two molecules PKD Protein Kinase D and Kidins220 Kinase D interacting substrate of 220 kDa also known as Ankyrin repeat rich membrane spanning or ARMS in order to define their role in neuronal processes and neuronal physiopathology We cloned Kidins220 as a novel neurospecific protein first substrate for PKD and this protein functions are now starting to be deciphered Kidins220 is an evolutionary conserved transmembrane protein with unique features which presents one single gene from C elegans to humans The distribution of Kidins220 in the nervous system its localization at the tip of extending neurites and its interaction with neurotrophin and ephrin receptors and several molecules we have identified in our laboratory suggest the participation of Kidins220 and PKD in neuronal differentiation survival and death and neurodegeneration We have recently found that Kidins220 undergoes a strong downregulation during neuronal excitotoxic death This type of neuronal death is involved in the development of several neurological syndromes and neurodegenerative diseases such as Parkinson Huntington or Alzheimer In this context we have discovered that Kidins220 positively contributes to neuronal survival and its absence makes neurons more vulnerable to excitotoxic damage In addition we have characterized the molecular mechanisms that contribute to decrease Kidins220 levels in excitotoxicity This information allowed us to design several strategies to prevent Kidins220 downregulation in order to protect neurons from neurodegenerative insults and increase their survival By investigating the role of Kidins220 in neuronal differentiation using primary hippocampal neurons in culture we have shown that it is a key regulator of neuronal polarity and dendrite development and maturation Furthermore we have found that this protein interacts and controls the activity of two microtubule regulatory proteins MAP1B and MAP2 which are key players in the regulation of these processes Given that these two molecules are present
