    patients using direct sequencing and next generation sequencing strategies Also we are characterizing the regulatory regions of all these genes employing EMSA and ChIP assay We are also studying the functions of FKRP and Fukutin whose implication in the O glycosylation process of alpha dystroglycan is still unknown For this we are using different constructions of these proteins mammalian cell based expression systems co immunoprecipitation assay and comparative glycoproteomics Functional studies of dystroglycanopathies using mouse animal model Rubio M Aza M Raducu M Tello A Rodríguez L Cruces J The use of animal models is essential to study the function of a gene and its associated pathology Our group has been focused on the study of POMT1 gene which encodes Protein O mannosyltransferase 1 the first enzyme participating in the synthesis of alpha dystroglycan s O mannosylglycans through which binds to extracellular matrix proteins such as laminin agrin neurexin pikachurin and perlecan Our laboratory was the first in generating a KO mouse model of one of the proteins involved in the synthesis of O mannosylglycans POMT1 PNAS 2004 However this KO mouse model was embryonic lethal due to defects in Reichert s membrane formation which is a basement membrane critical for embryo viability This is due to deficiencies in alpha dystroglycan glycosylation that is expressed in this membrane To resolve this issue our group is carrying our two strategies a Generation of chimeric mice using ES Pomt 1 whose contribution to the development of different tissues was significantly high Subsequently the resulting chimeras will be phenotyped using different strategies such as quantification of the degree of chimerism in each tissue type hearing tests retinography evaluation of motor functions MRI immunohistochemistry etc b Generation of Pomt1 conditional knockout mouse using Cre loxP system and analysis of POMT1 function in mouse general development and specifically in brain muscle and eye development Subsequently characterization and phenotyping of these mice will be performed using the aforementioned strategies Functional studies of dystroglycanopathies using chicken animal model Izquierdo A de Luís O Cruces J Coloma A Due to the limitations reported for the mouse system we are starting the generation of another animal model such as chicken which lacks Reichert s membrane and presents brain muscle and eye development similar to humans Our previous data reveal that the chicken ortholog of human POMT1 protein GgPomt1 is expressed in all chicken development stages with a peak in nervous system encephalon and spinal cord and also in eye and skeletal muscle These results suggest that chicken could be a suitable model system for the study of POMT1 function in alpha dystroglycan O mannosylation during embryonic development and also for the study of different dystroglycanopathies The previous use of morpholinos and or pRFPRNAi vectors for chicken embryos reinforces the feasibility of these studies Studies of alpha dystroglycan hypoglycosylation in cancer Rubio M Zimman H Cruces J Dystroglycan involvement in development progression and invasion of different cancers has been described a few years ago The interaction between the tumour cell
