    a the mechanisms by which PKC PKA abolish the induced effects by the Kvβ1 3 subnit b the amino acids involved inthese effects c the pharmacological consequences of the PKC PKA inhibition in Kv1 5 Kvβ1 3 channels and d the composition o the possible signaling complex of Kv1 5 Kvβ1 3 Pharmacological consequences of Kv1 5 Kvbeta1 3 interactions Macías A Cruz ADL Prieto A Moreno C González T Valenzuela C The outward potassium current I Kur is the main responsible of human atria repolarization and it is generated by the activation of Kv1 5 channels Its slow and incomplete inactivation and its voltage dependence explain its important role in maintaining the duration of the atrial action potentials This slow inactivation is modified when the α Kv1 5 subunits coassembly with the β subunits present in the human myocardium Kvβ1 3 and Kvβ2 1 The Kvβ2 1 subunit induces a higher degree of slow inactivation and a negative shift in the threshold of activation of the current On the other hand Kvβ1 3 induces an initial fast and partial inactivation a higher degree of slow inactivation and a negative shift in the threshold of activation of the current towards more negative membrane potentials The fast inactivation induced by the Kvβ1 3 subunit is produced by a similar mechanism to that described for local anesthetic and antiarrhythmic drugs Moreover Kv1 5 subunits coassembled with Kvβ1 3 subunits are 5 times less sensitive to bupivacaine than Kv1 5 subunits which suggests that the receptor site previously described for local anesthetics could be the natural receptor for the inactivating particle of Kvβ1 3 subunits Therefore we will study 1 the mechanisms by which Kvβ1 3 and Kvβ2 1 modify the slow inactivation of Kv1 5 channels and 2 the possible pharmacological consequences of the α β interactions using as pharmacological tools bupivacaine and a permanently charged bupivacaine analog N methyl bupivacaine Antiarrhythmic effects of omega 3 fatty acids from marine and plant origin Moreno C Valenzuela C There is a large body of experimental evidences that relate a diet rich in omega 3 fatty acids FA specially docosahexaenoic ecosapentaenoic and α linolenic acids DHA EPA and ALA respectively with 1 prevention of coronary disease 2 prevention of sudden death and specially 3 prevention of ventricular arrhythmias postmyocardial infarction The antiarrhythmic effects of omega 3 FA have been attributed to their capacity to modulate different ionic channels whose activity is involved in the genesis and or maintenance of cardiac action potentials AP In fact omega 3 FA block cardiac Na channels and interact with other ionic channels inhibiting I Kur I TO I Kr and I Ca and some of them i e DHA but not EPA increasing the magnitude of I Ks due to their effects on the accessory subunit KCNE1 Our main objectives are to determine 1 Whether omega 3 FA both marine and plant origen have an antiarrhythmic effect in patients with paroxysmal recurrent atrial fibrillation 2 The posible cardioprotective

    Reguera Research topics Transcriptional regulation of pancreatic function in the islets of Langerhans Our work is focussed on the study of the molecular mechanisms that regulate the development cell differentiation and function of pancreatic islets We are interested in the mechanisms of transcriptional regulation of gene expression and their implications for the etiopathogenesis of diabetes mellitus In addition we are interested in the study of the developmental alterations of the central nervous system related with gestational diabetes We have observed that the homeodomain transcription factor Alx3 is expressed in cells of the pancreatic islets of Langerhans but not in the exocrine pancreas Our data indicate that this factor is important for insulin glucokinase glucagon and somatostatin gene expression We have carried out detailed analyses of some of the regulatory elements located inthe promoter of these genes on which Alx3 acts as well as of interactions with other transcription factors that belong to the bHLH and homeodomain families Studies carried out using Alx3 deficient mice indicate that this factor is important for islet cell survival Thus beta cell mass in Alx3 deficient mice is markedly reduced as compared with control animals Alx3 null mice develop glucose intolerance and diabetes that worsens with age and triggers insulin resistance due to a defect in signalling in liver and muscle On the other hand our studies show that Alx3 is involved in the mechanisms that regulate neural tube closure in embryos Thus a proportion of Alx3 deficient embryos show cranial neural tube closure defects These defects correlate with an increase in the number of apoptotic cells in the cranial mesenchyme a region whose importance for the closure of the cranial neural tube is well documented Finally we have shown that the expression of Alx3 depends on the existence of adequate levels of folic acid
